Optimization of the tetrazolium dye (MTT) colorimetric
The MTT colorimetric assay is an established method of determining viable cell number in proliferation and cytotoxicity studies. This assay is based on the cleavage of the yellow tetrazolium salt, MTT, to form a soluble blue formazan product by mitochondrial enzymes, and the amount of formazan produced is directly proportional to the number of living, not dead cells, present during MTT exposure.

The development of the sulforhodamine B (SRB) protein staining assay for the in vitro measurement of cellular protein content of adherent and suspension cultures was established by Skehan et al. (1990); subsequently it was adopted for routine use in the National Cancer Institute in vitro antitumor screen (Kerkvliet, 1990; Monks et al., 1991). The dye binds to basic amino acids of cellular proteins and colorimetric evaluation provides an estimate of total protein mass which is related to cell ...

Results : Some recent papers describing the optimization of the MTT assay concern modifications in the final step and suggest using DMSO to improve formazan dissolution for absorbance readings 12.This requires separation of cells with bound formazan crystals from the MTT reaction medium.

The MTT assay is a colorimetric assay for assessing cell metabolic activity. NADH-dependent cellular oxidoreductase enzymes may, under defined conditions, reflect the number of viable cells present. These enzymes are capable of reducing the tetrazolium dye MTT 3--2,5-diphenyltetrazolium bromide to its insoluble formazan, which has a purple color. Other closely related tetrazolium dyes including XTT, MTS and the WSTs, are used in conjunction with the intermediate electron acceptor, 1-methoxy phen
